CD45.1/CD45.2 congenic markers induce a selective bias for CD8+ T cells during adoptive lymphocyte proliferation in lymphocytopenia mice

Abstract CD45.1/CD45.2 congenic markers have been frequently used to track hematopoietic lineage differentiation following stem and progenitor cell (HPSC) transplantation. However, several studies suggest that a bias exists in CD45.1 versus CD45.2 reconstitution from the transplanted HPSCs. Meanwhile, no definitive comparison has reported for mature immune cells as whether disparity can skew response. In this study using lymphocytopenia Rag1−/− mice hosts, we assessed proliferative potential of lymphocytes adoptive transfer T B harvested spleens mice. We found selective CD8+ showed significantly higher proliferation expression PD-1 compared hosts. This is likely due MHC-I restricted allogeneic stimulation CD4+ CD19+ contained same graft equivalent proliferation.
